DSS inhibits the activity of Taq polymerase. A) RNA from non-DSS-treated tissue was reverse transcribed, the cDNA was incubated in vitro with DSS, and the amplification of 36B4 was assessed by qPCR. Incubation with 0.01 g/L DSS triggered a small but significant decrease in the amplification of 36B4, and 0.05 g/L and higher concentrations of DSS fully inhibited the amplification of 36B4 by qPCR. B) Constant amounts of Taq polymerase (10 mU) were incubated with water or increasing concentration of DSS, and polymerase activity was assessed.
