Cell proliferation in response to stimulation with phytohemaglutinin (PHA). 2 × 105 PBMC were stimulated with different concentrations of PHA (final volume = 150 µl). After 48 h, reagent from the Quick Cell Proliferation Assay kit was added to each well and absorbance of the dye solution was measured at 450 nm after an additional 4 h of incubation. The graph shows the dose–response curve obtained for two individuals (a). 0.5–1.5 × 105 PBMC or 1 × 105 splenic mononuclear cells were stimulated with PHA (25 µg/ml; final volume = 150 µl) and proliferation was assessed after 48 h using the Quick Cell Proliferation Assay kit, as described in a. Work was performed in duplicates and data are expressed as mean ± SEM (b). Asterisks indicate a statistically significant difference (*p < 0.05 and ***p < 0.0005)
